I just returned today from my first visit!
You need to take the Hop On Hop Off Bus Tour because the city is so huge and you miss a lot when you just take the subway. They have a uptown tour which goes through Harlem, and a downtown tour which goes through Brooklyn. You can buy tickets online or from one of the many ticket agents around Times Square. You have to visit Times Square in the evening. You will feel like you're in the heart of New York, with all the people and all the stuff around you. And maybe visit a musical or a play on Broadway? And you should visit Empire State or Top of the Rock to get a view from the top.

Statue of Liberty, Staten Island Ferry, Co-op City, Katz's and Tiffany's
Central Park, Brooklyn Bridge, The Empire State (where Dylan lived) Coney Island and Times Square, Rockefeller Center - Wish I was there! (bonus points if you can name that song )

FAO Scwartz and all the other stores on 5th Ave. Plus just take a general walk through Central Park - Strawberry Fields, the Meadow, the Great Lawn, etc. Plus
all the other stuff everyone's named. Plus make a trip up to the Bronx to see the new Yankee Stadium and out to Queens for the new Mets Stadium, Citi
Field.

The bus tours are a good idea. It's super touristy but it will take you to all the must see places and let you see a lot of the city.
